RESUMEN

Bovine leptospirosis causes economic losses and raises public health concerns. It is possible that there are peculiarities in the epidemiology of leptospirosis in regions with a semiarid climate, such as the Caatinga biome in Brazil, where the climate is hot and dry, and the etiological agent require alternative routes of transmission. This study aimed to close knowledge gaps to the diagnosis and epidemiology of Leptospira spp. infection in cows from the Caatinga biome, Brazil. Samples of the blood, urinary tract (urine, bladder and kidney) and reproductive tract (vaginal fluid, uterus, uterine tube, ovary and placenta) were collected from 42 slaughtered cows. Diagnostic tests included were the microscopic agglutination test (MAT), polymerase chain reaction (PCR) and bacterial isolation. Anti-Leptospira spp. antibodies were found in 27 (64.3%) of the animals analyzed using MAT at a 1:50 dilution (cut-off 50), while 31 (73.8%) animals had at least one organ/fluid where the presence of Leptospira spp. DNA was identified, and 29 animals (69%) were positive at bacteriological culture. The highest sensitivity values for MAT were obtained at the cut-off point of 50. In conclusion, even under hot and dry climate conditions, it is possible that Leptospira spp. can spread through alternative routes such as venereal transmission; moreover, a cut-off of 50 is recommended for the serological diagnosis of cattle from the Caatinga biome.

Aimed with this study to evaluate vertical transmission of Neospora caninum in naturally infected sheep and to monitor the kinetics of antibodies against this protozoon in their lambs. Therefore, 48 pregnant ewes, from five herds, were divided into two groups: G1 - positive for anti-N. caninum antibodies, with 19 animals; and G2 - seronegative, with 29 animals. Blood samples were taken from the ewes and their lambs, immediately after birth, before ingesting colostrum, and 2, 7, 14, 21, 28, 35, 42, 49 and 56 days after birth. Analysis on serum antibodies was performed using the indirect immunofluorescent antibody test. Among the 19 seropositive mothers, six (31.6%) gave birth to lambs seropositive before ingesting colostrum and it was found that these lambs remained positive until the end of the study (56 days). Only one of the lambs, from a ewe that presented an antibody titer of 200, seroconverted after ingestion of colostrum. All the lambs that had been born from negative mothers remained negative throughout the experimental period. It was concluded that transplacental transmission was an important form of diffusion of N. caninum in the herds studied and that seropositive lambs maintained circulating antibodies during the period analyzed.

Resumo Objetivou-se avaliar a ocorrência da transmissão vertical de Neospora caninum em ovelhas naturalmente infectadas e monitorar a cinética de anticorpos para esse protozoário nos cordeiros. Portanto, foram selecionadas 48 matrizes prenhes, provenientes de cinco propriedades, e estas foram divididas em dois grupos: G1- 19 matrizes positivas para anticorpos anti-N. caninum antes da prenhez; e G2 - 29 matrizes soronegativas. Foram realizadas colheitas sanguíneas nas mães e cordeiros, no G1 e G2, imediatamente após a parição, antes da ingestão do colostro. Também foi possível realizar colheitas de sangue com 2, 7, 14, 21, 28, 35, 42, 49 e 56 dias após o nascimento. A pesquisa de anticorpos séricos foi realizada por meio da Reação de Imunofluorescência Indireta (RIFI). Das 19 matrizes soropositivas, seis (31,6%) pariram cordeiros soropositivos antes da ingestão do colostro, os quais mantiveram-se positivos até o final do experimento (56 dias). Apenas um dos cordeiros, filho de uma ovelha com título de anticorpos 200, soroconverteu após ingestão do colostro. Todos os cordeiros, filhos de mães negativas, mantiveram-se negativos durante todo o período experimental. Conclui-se que a transmissão transplacentária é uma importante forma de difusão do N. caninum nos rebanhos estudados e que os descendentes infectados, durante a gestação, mantiveram anticorpos circulantes durante o período analisado.

Brucellosis is one of the most common bacterial zoonoses worldwide affecting not only livestock and wildlife but also pets. Canine brucellosis is characterized by reproductive failure in dogs. Human Brucella canis infections are rarely reported but probably underestimated due to insufficient diagnostic surveillance. To improve diagnostics, we investigated dogs in a breeding kennel that showed clinical manifestations of brucellosis and revealed positive blood cultures. As an alternative to the time-consuming and hazardous classical identification procedures, a newly developed species-specific intact-cell matrix-assisted laser desorption/ionization-time of flight mass spectrometry analysis was applied, which allowed for rapid identification of B. canis and differentiation from closely related B. suis biovar 1. High-throughput sequencing and comparative genomics using single nucleotide polymorphism analysis clustered our isolates together with canine and human strains from various Central and South American countries in a distinct sub-lineage. Hence, molecular epidemiology clearly defined the outbreak cluster and demonstrated the endemic situation in South America. Our study illustrates that MALDI-TOF MS analysis using a validated in-house reference database facilitates rapid B. canis identification at species level. Additional whole genome sequencing provides more detailed outbreak information and leads to a deeper understanding of the epidemiology of canine brucellosis.

Leptospirosis is a globally distributed disease associated with reproductive failures in livestock; however, its pathogenesis has not been fully elucidated. Results from the present study indicate there is a presence of Leptospira sp. in organs and fluids of fetuses from ewes slaughtered in the semiarid region of Brazil. Twenty-nine fetuses from 23 ewes determined to be Leptospira sp.-positive using PCR were sampled (14 and 15 in dry and rainy seasons, respectively). Fetal samples of blood, central nervous system (CNS), lung, liver, spleen, stomach contents, peritoneal fluid, kidney, bladder, urine and reproductive system were collected. Diagnostic methods included the microscopic agglutination test (MAT), polymerase chain reaction (PCR) and bacterial isolation. Of the 29 fetuses, 24 (82.8 %) had at least one Leptospira sp.-positive organ or fluid, as determined using PCR, and of a total of 209 samples, 62 (29.7 %) contained leptospiral DNA. Of the 99 samples collected during the dry season, 42 (42.4 %) were positive, and of 110 samples collected during the rainy season, 20 (18.2 %) were positive (P = 0.0001). There was deoxyribonucleic acid (DNA) sequencing of three samples of kidney, CNS and liver, and in all of these, there was 99.3 % similarity with Leptospira interrogans. Leptospires were present in cultures of pooled samples from fetuses with deformities. Results indicate there is vertical (maternal-to-fetus) transmission which would represent an alternative transmission route for the spread of Leptospira sp. in ewes, suggesting molecular detection is essential in the investigation of leptospirosis in fetuses to identify animals that have been infected with this bacterium.

It is possible that there are peculiarities in the epidemiology of leptospirosis in regions with a semiarid climate, where the environment is often adverse, allowing the occurrence of alternative transmission routes. The objective of the work was to generate contributions to the diagnosis and epidemiology of Leptospira sp. infection in sheep reared in semiarid conditions, using serological, molecular and microbiological techniques for diagnosis in dry and rainy seasons. Samples of blood, vaginal fluid, urine, bladder, kidney, uterus, uterine tube, ovary and placenta were collected from 104 sheep (52 animals per season - dry and rainy) slaughtered in the Brazilian semiarid. Diagnostic tests performed were microscopic agglutination test (MAT), polymerase chain reaction (PCR) and bacterial isolation. Anti-Leptospira sp. antibodies were found in 26 (25%) of the animals analyzed by MAT at 1:50 dilution (cut-off 50), while 69 (66.3%) animals had at least one organ/fluid with the presence of Leptospira sp. DNA. Overall, PCR was performed on 758 fragments of organs/fluids from the genital and urinary tracts, and 519 (68.5%) samples tested positive. PCR-positivity was statistically different in dry (46.2%) and rainy (11.5%) seasons for vaginal fluid. It was possible to perform the DNA sequencing in nine samples with 99% similarity to L. interrogans and recovery of viable strains in three samples of vaginal fluid. Regardless of the biological material used in PCR to detect carrier animals and the season, the highest MAT sensitivity values were obtained with cut-off 50 compared to 100. The results obtained indicate that, even in the adverse environmental conditions of the semiarid region, leptospires may survive and propagate by alternative routes of transmission, such as sexual, and the presence of PCR-positive genital tracts in ewes suggests that sexual transmission may play an important role in the epidemiology of the disease in sheep in Brazilian semiarid. In addition, it is suggested the use of titer 50 as cut-off point at serology in semiarid conditions.

Os espaços coletivos com grande circulação de pessoas e animais tutelados ou errantes, como as praças públicas, podem gerar um problema para a saúde pública, uma vez que os animais podem eliminar fezes contaminadas por parasitos nesses locais, desempenhando um importante papel epidemiológico na transmissão de zoonoses parasitárias. Portanto, este trabalho teve como objetivo avaliar a contaminação por parasitos zoonóticos em dez praças públicas da cidade de Sousa, Paraíba, bem como avaliar o melhor método de diagnóstico de infestação ambiental por estes parasitas. Foram feitas coletas de solo em cinco pontos diferentes de cada praça, por raspagem superficial em quadrantes delimitados por 15 cm, sendo coletados 200g da parte central de cada ponto. Os métodos de diagnósticos utilizados foram Flutuação Simples, Sedimentação Simples, Faust e Centrífugo-Flutuação em Solução de Sacarose. Das praças avaliadas, 90% (9/10) obtiveram pelo menos uma amostra positiva. O gênero de parasito mais frequente foi Ancylostoma spp., presente em 70% (7/ 10) das praças analisadas; seguido por Strongyloides spp., coccídeos e Taenia spp., em 40% (4/10); Toxocara spp., em 30% (3/10); e Cystoisospora spp. e Trichuris spp., em 10% (1/10). O método de Sedimentação Simples foi o mais eficaz, resultando em 44% (22/50) de amostras positivas. Os demais métodos não atingiram 15% de positividade. Concluiu-se que as praças públicas analisadas encontram-se altamente infestadas por parasitos zoonóticos, representando um risco a saúde pública no município de Sousa-PB.

Abstract Occurrence of infection or exposure to Ehrlichia canis, Hepatozoon canis and Rickettsia spp. was detected in feral cats living in two fragments from Atlantic rainforest, in Natal, RN, Brazil, and in dogs living around the parks. While serum samples were collected from 155 animals (53 cats living in the parks; 29 dogs living in human homes around the parks; and 73 dogs living at an animal control center - ACC), spleen samples were collected from 20 dogs that were euthanized at ACC. Serum samples were analyzed to Rickettsia spp. and E. canis antibodies using the indirect immunofluorescence assay. Seventeen of the 102 dogs (17%) had E. canis antibodies and 13% (20/155) of all dogs and cats (i.e. 3% (3/102) of the dogs and 32% (17/53) of the cats) were seropositive for Rickettsia spp. antigens. The animals were therefore been exposed to R. amblyommatis or by a very closely related genotype. Among the 20 dog spleen samples analyzed, eight were PCR positive for E. canis and two for H. canis (GenBank accession number MG772657 and MG772658, respectively). In none of the spleen samples were obtained amplicons for Babesia spp. through PCR. This study provided the first evidence that Rickettsia of the spotted fever group is circulating among dogs and cats in Natal.


Resumo A ocorrência de infecção ou exposição para Ehrlichia canis, Hepatozoon canis e Rickettsia spp. foi determinada em gatos ferais que viviam em dois fragmentos da Mata Atlântica, localizados em Natal, RN, Brasil e em cães que viviam em torno dos parques e em outras regiões da cidade. Enquanto amostras de soro foram coletadas de 155 animais (53 gatos que viviam nos parques, 29 cães com domicilio em torno dos parques e 73 cães do Centro de Controle de Animais -CCA), fragmentos de baço foram coletados de 20 cães eutanasiados no CCA. A detecção de anticorpos nas amostras de soros coletadas contra Rickettsia spp. e E. canis foi realizada pela Reação de Imunofluorescência Indireta. Dezessete dos 102 cães (17%) apresentaram anticorpos anti E. canis e 13% (20/155) de todos os cães e gatos (ou seja, 3% (3/102) dos cães e 32% (17/53) dos gatos) foram soropositivos para antígenos de Rickettsia spp. Os animais foram considerados expostos à R. amblyommatis ou a um genótipo muito relacionado. Entre as 20 amostras de baço de cães analisadas, oito foram positivas para E. canis e duas para Hepatozoon canis (números de acesso ao Genbank MG772657 e MG772658, respectivamente). Nenhuma das amostras de baço produziram amplicons de Babesia spp. na PCR. Observou-se, pela primeira vez, a circulação de Rickettsia do grupo da febre maculosa em cães e gatos em Natal, RN.

This study evaluated the protective effects of resveratrol on the prostate development of rats exposed to TCDD. Pregnant rats received TCDD (1 µg/kg) at GD15 and/or RES (20 mg/kg/day) from GD10 to PND21. Newborn and adult males from Control, TCDD, TCDD + RES and RES groups were euthanized and the prostate was excised. On PND1, there was a reduction in the number of prostatic buds, AR-positive mesenchymal cells and proliferation index in epithelial and mesenchymal cells in TCDD group, but restored by RES. AhR immunoreactivity was greater in TCDD group than the other groups. On PND90, there was higher frequency of functional hyperplasia in the distal area of the prostate acini in TCDD group, but restored by RES. AhRR expression was higher in the TCDD while NRF2 was higher in the TCDD + RES compared to the other groups. Resveratrol was able to reduce the adverse effects of TCDD on prostate development and its long-term repercussions.

Occurrence of infection or exposure to Ehrlichia canis, Hepatozoon canis and Rickettsia spp. was detected in feral cats living in two fragments from Atlantic rainforest, in Natal, RN, Brazil, and in dogs living around the parks. While serum samples were collected from 155 animals (53 cats living in the parks; 29 dogs living in human homes around the parks; and 73 dogs living at an animal control center - ACC), spleen samples were collected from 20 dogs that were euthanized at ACC. Serum samples were analyzed to Rickettsia spp. and E. canis antibodies using the indirect immunofluorescence assay. Seventeen of the 102 dogs (17%) had E. canis antibodies and 13% (20/155) of all dogs and cats (i.e. 3% (3/102) of the dogs and 32% (17/53) of the cats) were seropositive for Rickettsia spp. antigens. The animals were therefore been exposed to R. amblyommatis or by a very closely related genotype. Among the 20 dog spleen samples analyzed, eight were PCR positive for E. canis and two for H. canis (GenBank accession number MG772657 and MG772658, respectively). In none of the spleen samples were obtained amplicons for Babesia spp. through PCR. This study provided the first evidence that Rickettsia of the spotted fever group is circulating among dogs and cats in Natal.

Canine brucellosis is caused by Brucella canis, a gram negative and facultative intracellular bacterium that is commonly associated with reproductive failures in dogs. The accurate diagnosis of the infection relies on the use of serological tests associated with blood culturing to guarantee sensitivity. The polymerase chain reaction (PCR) can replace the culturing procedure for the direct diagnosis of the infection because of its speed, high specificity and sensitivity values; however, it depends on some laboratory infrastructure to be conducted. The loop-mediated isothermal amplification (LAMP) may be an alternative method for DNA amplification in a shorter period, using simpler equipment, and with a lower cost. This study evaluated the potential of molecular tools based on PCR and LAMP using primers targeting the insertion sequence IS711 for Brucella detection in three groups of dogs (infected, non-infected and suspected of brucellosis), which were determined according to the results of blood culturing and clinical examination. The performance of the three diagnostic tests was also determined using McNemar test and Kappa coefficient. The proportion of positive samples detected by blood culturing, PCR and LAMP was respectively 31.57% (18/57), 33.34% (19/57), and 14.03% (8/57). The agreement between blood culturing and PCR was almost perfect, while the agreement of PCR and blood culturing compared to LAMP was fair. The diagnostic sensitivity of PCR and LAMP was respectively 100% (18/18) and 44.44% (8/18), while the diagnostic specificity of both tests was 100% (21/21). LAMP performance was not satisfactory for canine brucellosis diagnosis because of the low diagnostic sensitivity of the test. The IS711 based PCR, otherwise, showed high values of sensitivity and specificity, which makes it a good alternative for use for the rapid diagnosis of canine brucellosis.

INTRODUCTION:: Leishmaniasis is endemic to the Northern, Northeastern, Central-Western, and Southeastern regions of Brazil. We aimed to assess the epidemiological situation of leishmaniasis in humans and dogs in indigenous villages located in the States of Mato Grosso and Tocantins using a serological survey conducted in May 2011. METHODS:: Serum samples were collected from 470 humans and 327 dogs living in villages of the Urubu Branco and Tapirapé Karajá indigenous reserves. The samples were analyzed for the presence of Leishmania spp. antibodies using the indirect fluorescent antibody test (IFAT), enzyme-linked immunosorbent assay (ELISA) with a crude antigen (CA) and soluble antigen (SA), and Dual Path Platform (DPP®) immunoassay for canine visceral leishmaniasis. RESULTS:: Of 470 human samples tested, two (0.4%) were positive using IFAT. Among 327 dog samples tested, 28 (8.6%) were positive using ELISA CA, five (1.5%) using ELISA SA, two (0.6%) using IFAT, and none using DPP® immunoassay with Leishmania infantum chagasi antigen. When Leishmania amazonensis antigen was used, 20 (6.1%) samples were positive using ELISA CA and four (1.2%) using IFAT. CONCLUSIONS:: There was a low prevalence of infection in the region, and significant differences among the main serological methods used for the diagnosis of leishmaniasis. These findings indicated that the detection of Leishmania spp. requires further study and improvement.

Prostate morphogenesis is regulated by androgens hormones and modulated by morphogenetic proteins such as Bone Morphogenetic Proteins (BMPs). This study aims to investigate the effects on prostate development in male offspring and differentiation after gestational and lactational maternal exposure to Di-n-butyl-phthalate (DBP), an important environmental contamination. Pregnant Wistar rats received 100 or 500mg/kg of DBP (DBP100 and DBP500), by gavage, from gestation day 15 (GD15) until postnatal day 21 (PND21). The pups were euthanized on PND1 and PND21. Anogenital distance and testosterone levels decreased in animals from exposed mothers (DBP100 and 500) on PND1. A three-dimensional reconstruction model of the prostatic urethra showed reduction in the prostatic buds in the DBP500 group. AR expression and α-actin immunoreactivity decreased, and BMP-4 expression was lower on PND1 for DBP500. These results showed that DBP exposure, especially at a higher dose, delayed prostate morphogenesis by reducing the testosterone/AR axis and BMP-4 expression.

Resumo A ocorrência de infecção ou exposição para Ehrlichia canis, Hepatozoon canis e Rickettsia spp. foi determinada em gatos ferais que viviam em dois fragmentos da Mata Atlântica, localizados em Natal, RN, Brasil e em cães que viviam em torno dos parques e em outras regiões da cidade. Enquanto amostras de soro foram coletadas de 155 animais (53 gatos que viviam nos parques, 29 cães com domicilio em torno dos parques e 73 cães do Centro de Controle de Animais -CCA), fragmentos de baço foram coletados de 20 cães eutanasiados no CCA. A detecção de anticorpos nas amostras de soros coletadas contra Rickettsia spp. e E. canis foi realizada pela Reação de Imunofluorescência Indireta. Dezessete dos 102 cães (17%) apresentaram anticorpos anti E. canis e 13% (20/155) de todos os cães e gatos (ou seja, 3% (3/102) dos cães e 32% (17/53) dos gatos) foram soropositivos para antígenos de Rickettsia spp. Os animais foram considerados expostos à R. amblyommatis ou a um genótipo muito relacionado. Entre as 20 amostras de baço de cães analisadas, oito foram positivas para E. canis e duas para Hepatozoon canis (números de acesso ao Genbank MG772657 e MG772658, respectivamente). Nenhuma das amostras de baço produziram amplicons de Babesia spp. na PCR. Observou-se, pela primeira vez, a circulação de Rickettsia do grupo da febre maculosa em cães e gatos em Natal, RN.
